Experimental Studies on the Mechanism of Leukocytosis Induced by Water Soluble Tissue Extracts

Abstract
Leukocytosis producing effect was tested on normal rabbits and those which had cervical cord section at the lower level or slpanchnicectomy by injection with saline extract of emulsion of the heart, liver, spleen, pancreas, kidney or adrenal gland and autolized tissue of the liver, spleen or kidney obtained from normal rabbits.
And, furthermore, same test was made on normal rabbits by injection of sera obtained from the normal rabbits and those with lower cervical cord section which were injected with such extracts.
These saline extracts and sera were injected intravenously into the rabbits and the following conclusions were obtained.
l. The saline extracts of the normal and autolized tissues have the ability of inducing pseudoeosinophilia with nuclear shift to the left.
2. The leukocytosis-producing effect of the normal or autolized tissue extracts can also be proved in the animals with lower cervical cord section or bilateral splanchnicectomy.
3. It is reasonable to conclude that there appears in the circulating blood a certain endogeneous leukocytosis producing substance which is different from the tissue extract injected previously, when the normal or autolized tissue extracts are injected into normal rabbits and those with lower cervical cord section.
4. The leukocytosis due to normal or autolized tissue extracts is the phenomenon caused by releasing the endogeneous leukocytosis-producing substance secondarily in the circulating blood, in consequence of stimulating or injuring a certain living tissue by the active substance contained in such tissue extracts. Thus, the mechanism of the leukocytosis is not influenced by leukocyte-regulating center, I believe,
5. As far as the concepts of pathogenesis of neutrophilia associated with inflammatory or non-inflammatory pathologic processes are concerned, the following opinions must be considered on the basis of the above conclusions.
A. The primary leukocytosis-producing substance which could be released from the injured tissues into the circulating blood stimulates or injures a certain living tissue without the interference of the leukocyteregulating center and produces the secondary leukocytosis-producing substance.
B. This substance enters the circulating blood, acting directly on the bone marrow and induces the mobilization of neutrophils.
